I'd say the chances of this happening are very good. Especially if you rent from an airport that is extremely busy. They are more interested in selling you that upgrade, fuel purchase option, or insurance. It will generally be the independent city locations that will have a closer look at the pricing structure.
In my experiences where I had to surrender coupons, DBCs, upgrades, vouchers they usually just look at the expiry date.
